C1 Quick setup Guide
1. Open 4 windows in program. In "view" Tab force and then unlock each window.
2. Configure > Confocal > Grabber Tab. Check linear gain box (makes gain display from 1-10). Check pixel time correction box (adjusts gain as dwell time is increased).
3. System has no dark level control. Instead, the black level is custom adjusted for different PMT amplitudes. Avoids customer throwing away low signal sample when trying to adjust an 8-bit output display. Configure >Confocal >Grabber Tab >adjust. Follow directions for scope. For first gain setting raise PMT voltage until noise just visible for each laser. Accept and do next level, and so on.
4. check that good signal is achieved with PMT readout under 6 for all lasers. Note that the standard Pollen Grain has poor 488nm response. Use green slide instead. Argon laser should be in standby mode (3-4 mw) with ND4 filter.
5. Set focus home, top (clockwise) and bottom (counter-clockwise). Step size will be suggested. Accept or over-ride. Gather data stack. and SAVE IMMEDIATELY!
6.Rock display stack . Data > Volume > Render > Enable. color = best. 21 5-degree steps starting at -30 degrees. Save as AVI file. save option 10 frames /sec.
7. Note that all files are saved in non-proprietary .IDS format. This is the only true 12-bit public domain format available. Photoshop, Autoquant, and Metamorph are some of the programs that read this format. .ICS is the file hheader and is always generated when an .IDS file is saved. Other companies convert to 8-bit TIFF files or uses RAW format that has no header information.
