How to segment and measure Channels with Consistent numbering: 1) Channel 1 + Channel 2 2) Channel 1 3) Channel 2 4) Background 5) Channel 1 – Background
1. Channel 1 + Channel 2Segment objects using a multi or any single channel image. In example below, 2-Channel Image was segmented using the Blue channel. Cell #1 is highlighted in magenta and has an average grey scale value of 144.47. A binary image (and cell numbering mask) is automatically generated with the measurement and is saved by the menu command REFERENCE > Binary -> Reference binary. 2. Channel 1 (Blue) The cell numbering g binary mask is applied to the Blue channel by selecting the blue channel and using the menu command REFERENCE > Binary <- Reference binary. Note the average value for cell #1 is 191.19.
3. Channel 2 (Red) The cell numbering g binary mask is applied to the Red channel image by selecting the image Red Channel image and using the menu command REFERENCE > Binary <- Reference binary. Note the average value for cell #1 is about 97. Note that the value 144 is the average of the values 191 and 97.
4. Background Measurement Using a region of interest ROI tool select an area of the background. In the example below, the average background is 8.03.
